  Abstract: This paper systematically reviews the biological functions, mechanisms of action, and clinical translation potential of neuropeptide S (NPS). NPS exerts diverse physiological regulatory effects by binding to the G protein-coupled receptor NPSR and activating downstream signaling pathways. Moreover, NPSR gene polymorphisms correlate closely with susceptibility to various psychiatric disorders. NPS plays a pivotal role in emotional regulation, sleep-wake modulation, analgesia, and cognitive maintenance, and it also regulates anesthesia emergence, gastrointestinal feeding behavior, and psychiatric diseases.
