Abstract: Nogo-A/B is an important member of the reticulon family 4 (RTN4) and is widely expressed in the central nervous system and peripheral tissues. Historically, Nogo-A/B was regarded as a key inhibitory factor of axonal regeneration in the central nervous system. In recent years, growing evidence has shown that Nogo-A/B regulates macrophage activation and function and participates in inflammatory responses. This article is a narrative review that focuses on in vivo and in vitro evidence from the past 5–7 years, supplemented by classical literature. Centering on the interactions between Nogo-A/B and macrophages, we summarize evidence for its roles in the stages of recruitment, migration, polarization, and effector functions, and outline the potential mechanisms across multiple inflammation-related diseases. These lines of evidence help to elucidate the occurrence and development of inflammatory diseases and provide avenues for targeted therapy.
