Research Progress on the Association between the Calsyntenin Family and Diseases

    Abstract: Calsyntenin (CLSTN) is an important member of the cadherin superfamily (CDH), characterized by cadherin-related domains, transmembrane domains, and intracellular domains, playing crucial roles in cell adhesion, signal transduction, and vesicle transport. Recent studies have revealed that CLSTN family members are closely associated with the development of various diseases, including neurological disorders, cardiovascular diseases, and cancers. CLSTN1 primarily participates in Alzheimers disease pathology by regulating amyloid precursor protein (APP) metabolism and plays a role in myocardial repair after myocardial infarction. CLSTN2 is associated with synaptic plasticity regulation and cardiac arrhythmias. CLSTN3 is involved in lipid metabolism regulation and exhibits abnormal expression in multiple tumors. In-depth investigation of the roles of CLSTN family members in different diseases may provide novel targets and strategies for disease diagnosis, treatment, and drug development. This review systematically summarizes the structural and functional characteristics of CLSTN family members and their mechanisms of action in various diseases.
