The Role of Histone Lactylation in Neurological Disorders: Progress and Challenges

Abstract: Since its discovery in 2019, histone lactylation has emerged as a research hotspot in neurological disorders. This review systematically summarizes recent advances in its roles across neurodegenerative diseases (such as Alzheimer‘s disease and Parkinson’s disease), acute brain injuries (including ischemic stroke and subarachnoid hemorrhage), as well as neurodevelopmental and metabolism-related disorders. Based on this, we further explore the multidimensional regulatory patterns and common mechanisms of lactylation modifications and discuss the potential of targeting lactylation pathways as therapeutic strategies.
