Hirudin targeting mTOR signaling in asthma:From molecular mechanisms to clinical applications

    Abstract: As a natural thrombin inhibitor, hirudin has recently been demonstrated to regulate autophagy, cell proliferation, and inflammatory responses through modulation of the mammalian target of rapamycin (mTOR) signaling pathway. The mTOR pathway plays a pivotal role in immune regulation, airway remodeling, and mucus secretion in asthma. This review integrates the mechanisms of the hirudin-mTOR axis observed in glioma, renal fibrosis, and peripheral nerve injury, combined with the dual roles of mTOR in Group 2 innate lymphoid cell (ILC2) activation, airway inflammation, and autophagy regulation, to explore the theoretical basis for hirudin as a potential anti-asthmatic agent, providing novel insights into expanding its clinical applications.
