    Abstract: Depression is a very complicated and multifactoral mental illness, it’s cause has yet to be discovered. There are also other mechanisms being studied like cell stress, neuroinflammation and impaired neuroplasticity along with neurotransmitters themselves. As a classical tumor suppressor gene,TP53 can encode the p53 protein, it’s a important key regulator of cellular stress response which is related with lots kinds of biological process like cell cycling, apoptosis and metabolism reprogramming. This article summarizes and reviews the latest research regarding role of p53 signaling pathway in depression’s occurrence development systematically reviewed on how may be mechanism tp53/p53signaling path affect developement on depressive behavior throw neuro inflamatory process,oxidative stress,mitochondrial function ,neural proliferation,synaptic plasticity also discussed what might feasible and difficult with this kind new therapeutic approach targeting to tp53/p53 signaling path way .
