Abstract: Objective: To retrospectively analyze the efficacy and safety of preoperative chemotherapy of two or three drugs in patients with locally advanced gastric/gastroesophageal adenocarcinoma in the real world. Methods: Retrospective collection of cT3-4N+M0 gastric/gastroesophageal adenocarcinoma admitted to the Affiliated Cancer Hospital of Xinjiang Medical University between January 2016 and December 2021 and underwent surgery after neoadjuvant chemotherapy. According to the exclusion criteria, a total of 112 cases were collected, including 36 cases in the two drug group, 76 cases in the three-drug group. All patients underwent preoperative chemotherapy and postoperative adjuvant treatment. Results: There is no significant difference between the two-drug/three-drug regimen for the patients OS, DFS, R0 rate, tumor downgrade(P>0.05); the patients age, surgical method, tumor downgrade, postoperative pathological staging, and pathological differentiation type are the independent risk factors affecting the patients OS and DFS (P<0.05). Conclusion: The preoperative chemotherapy regimen of two or three drugs has no significant impact on the prognosis of the patient, the decline of the tumor, and the resection of R0/R1, but the patients age, postoperative pathological staging, the downgrade of the tumor, and the pathological differentiation type have a certain impact on the prognosis of the patient.
