摘要:目的 针对咳嗽变异性哮喘(Cough Variant Asthma, CVA)现有疗法的局限性,本研究结合网络药理学预测与动物实验验证,系统探索小柴胡汤(Xiaochaihu Decoction, XCHD)治疗CVA的作用机制。方法 利用数据库筛选XCHD活性成分及其潜在靶点、CVA相关疾病靶点、构建“药物-成分-疾病-靶点”互作网络,并进行相关通路富集分析。建立卵清蛋白(Ovalbumin, OVA)诱导的CVA小鼠模型,评价XCHD对肺组织病理学及血清炎症因子水平的影响。结果 筛选出XCHD潜在活性成分198个,药物-疾病共同靶点212个。核心活性成分可能通过干预辅助性T细胞17(T helper 17 cell, Th17)分化、白细胞介素-17(Interleukin-17, IL-17)等信号通路发挥作用。动物实验表明,XCHD显著减轻肺部炎症细胞浸润和组织损伤,并显著降低血清IL-6、IL-17和免疫球蛋白E(Immunoglobulin E, IgE)水平(P均<0.05)。结论 XCHD治疗CVA具有“多成分-多靶点-多通路”协同作用的特点,其机制可能涉及调控IL-6/信号转导和转录激活因子3(Signal Transducer and Activator of Transcription 3, STAT3)及IL-17等信号通路,减轻气道炎症和降低IgE水平
Abstract: Objective Given the limitations of existing therapies for cough variant asthma (CVA), this study systematically explored the mechanisms of Xiaaihu Decoction (XCHD) in the treatment of CVA by combining network pharmacology prediction and animal experiment validation. Methods The active ingredients and potential targets of XD were screened using databases, as well as the targets of CVA-related diseases. A protein interaction network of drug-component-disease-target was constructed, and pathway enrichment analysis was performed. Ovalbumin (OVA)-induced CVA mouse model was established to evaluate the effects of XCHD on pathological changes in lung tissue and the levels of serum inflammatory factors Results A total of 198 potential active ingredients and 212 common drug-disease targets were screened. Core active ingredients may play a role through the intervention of helper 17 cell (Th17) differentiation, interleukin-17 (IL-17) and other signaling pathways. Animal experiments showed that XCH significantly reduced the infiltration of inflammatory cells and tissue damage in the lungs and significantly reduced the levels of serum IL-6, IL-17 and Immunoglobulin E (IgE) (P < 0.05). Conclusion XCHD has the characteristics of multiple components-multiple targets-multiple pathways syneristic effects in the treatment of CVA. Its mechanism may involve regulating IL-6/signal transducer and activator of transcription 3 (STAT3) and IL-7 signaling pathways, reducing airway inflammation and lowering IgE levels.
