Effects of Qingre Antai Gel on ANXA1/FPR2 Axis and Cell Apoptosis in Gingival Tissue of Rats with Pregnancy Gingivitis

• 摘要：要 目的：基于ANXA1/FPR2轴探究自拟清热安胎凝胶对实验性妊娠期龈炎大鼠的干预效果及作用机制。方法：采用缝线结扎法构建SD大鼠妊娠期龈炎模型，随机分为4组：空白对照组、模型组、阳性对照组（生理盐水冲洗）、中药组（清热安胎凝胶）。干预7 天后，观察大鼠牙龈状态，qPCR检测牙龈组织中膜联蛋白1（Annexin A1，ANXA1）、甲酰肽受体 2（FPR2）基因表达，Western blot检测牙龈组织ANXA1、FPR2蛋白表达，TUNEL荧光染色观察牙龈细胞凋亡水平。结果：与空白组相比，模型组大鼠牙龈红肿明显，ANXA1、FPR2 mRNA表达量分别上升至空白组的2.64倍和2.06倍，蛋白表达代偿性轻度升高，牙龈凋亡细胞大量增多；阳性组可减轻牙龈红肿，ANXA1、FPR2 mRNA表达量显著上调至空白组的3.62倍和3倍，蛋白表达量也大幅升高，凋亡细胞数量少于模型组。中药凝胶组可显著减轻牙龈红肿，且ANXA1、FPR2 mRNA表达量显著上调至空白组的5.31倍和4.51倍，蛋白表达量也大幅升高，凋亡细胞数量少于阳性对照组。结论：清热安胎凝胶可上调妊娠期龈炎大鼠牙龈组织ANXA1及FPR2表达，并降低组织细胞凋亡水平，减轻牙龈组织红肿及损伤，缓解妊娠期龈炎局部损伤，为孕期牙周局部安全用药提供实验依据。

  Abstract: Objective: To investigate the intervention effect and mechanism of self-made Qingre Antai gel on experimental pregnancy gingivitis rats based on ANXA1/FPR2 axis. Methods: The pregnancy gingivitis model in SD rats was established by suture ligation method. The rats were randomly divided into four groups: blank control group, model group, positive control group (normal saline irrigation), and Chinese medicine group (Qingre Antai gel). After 7 days of intervention, the gingival status of rats was observed. The gene expressions of ANXA1 and FPR2 in gingival tissue were detected by qPCR, the protein expressions of ANXA1 and FPR2 were detected by Western blot, and the level of gingival cell apoptosis was observed by TUNEL fluorescence staining. Results: Compared with the blank group, the model group showed obvious gingival redness and swelling, the mRNA expression levels of ANXA1 and FPR2 increased to 2.64 and 2.06 times of the blank group respectively, the protein expression increased compensatorily, and the apoptotic cells in gingiva increased significantly. The positive control group could alleviate gingival redness and swelling, the mRNA expression levels of ANXA1 and FPR2 increased significantly to 3.62 and 3.00 times of the blank group respectively, the protein expression increased significantly, and the number of apoptotic cells was less than that in the model group. The Chinese medicine gel group could significantly alleviate gingival redness and swelling, and the mRNA expression levels of ANXA1 and FPR2 increased significantly to 5.31 and 4.51 times of the blank group respectively, the protein expression also increased significantly, and the number of apoptotic cells was less than that in the positive control group. The differences among groups were statistically significant (P<0.05). Conclusion: Qingre Antai gel can up-regulate the expressions of ANXA1 and FPR2 in gingival tissue of pregnancy gingivitis rats, reduce the level of tissue cell apoptosis, alleviate gingival tissue redness, swelling and damage, and relieve local damage of pregnancy gingivitis, providing experimental basis for safe local medication during pregnancy.
