摘要:目的 本研究旨在基于UPLC-Q-Orbitrap-MS技术与网络药理学,系统解析孟河医派特色药对“核桃肉炒补骨脂”治疗肾阳虚型抑郁症的化学成分及其潜在作用机制。方法 采用UPLC-Q-Orbitrap-MS技术对核桃肉炒补骨脂提取物进行化学成分鉴定;通过网络药理学方法筛选活性成分及其作用靶点,构建“成分-靶点-通路”网络,并通过GO与KEGG富集分析探讨其治疗抑郁症的生物学过程与信号通路。结果 从核桃肉炒补骨脂中鉴定出49种化学成分,包括补骨脂素、异补骨脂素、补骨脂二氢黄酮甲醚等14个明确化合物。网络药理学筛选出30个活性成分、117个交集靶点、20条关键通路,涉及神经活性配体-受体相互作用、cAMP信号通路、5-羟色胺能突触、PI3K-Akt信号通路等。关键成分包括新补骨脂异黄酮、补骨脂素、异补骨脂素等。结论 核桃肉炒补骨脂可能通过多成分、多靶点、多通路协同作用,调节神经递质、抗炎、抗氧化及神经保护等机制,从而发挥治疗肾阳虚型抑郁症的作用,为后续实验验证与临床应用提供理论依据。
Abstract: ABSTRACT:Objective This study aimed to systematically investigate the chemical constituents and potential mechanism of the characteristic herb pair Juglandis Semen fried with Psoraleae Fructus from the Menghe Medical School in treating kidney-yang deficiency type depression, based on UPLC-Q-Orbitrap-MS technology and network pharmacology. Method The chemical components of the extract from Juglandis Semen fried with Psoraleae Fructus were identified by UPLC-Q-Orbitrap-MS. Active components and their potential targets were screened using network pharmacology methods. A component-target-pathway network was constructed, and the biological processes and signaling pathways involved in its antidepressant effects were explored through GO and KEGG enrichment analysis.Results A total of 49 chemical components were identified from Juglandis Semen fried with Psoraleae Fructus, including 14 confirmed compounds such as psoralen, isopsoralen, and bavachinin. Network pharmacology screening identified 30 active components, 117 intersection targets, and 20 key pathways, involving neuroactive ligand-receptor interaction, cAMP signaling pathway, serotonergic synapse, PI3K-Akt signaling pathway, among others. Key active components included neobavaisoflavone, psoralen, isopsoralen, etc.Conclusion Juglandis Semen fried with Psoraleae Fructus may exert therapeutic effects on kidney-yang deficiency type depression through multi-component, multi-target, and multi-pathway synergistic actions, potentially involving mechanisms such as neurotransmitter regulation, anti-inflammation, anti-oxidation, and neuroprotection. This study provides a theoretical basis for subsequent experimental validation and clinical application.
