Molecular mechanisms and therapeutic targets of ferroptosis in oral diseases

    Abstract: Ferroptosis is a form of regulated cell death (RCD) driven by iron-dependent lipid peroxidation. Recent studies have found that ferroptosis plays an important role in the pathological process of oral diseases. This article systematically elaborates on the molecular regulatory mechanisms of ferroptosis in various oral diseases such as oral squamous cell carcinoma, periodontitis, and temporomandibular joint osteoarthritis, and deeply explores the role of key targets such as glutathione peroxidase 4 (GPX4) and acyl-CoA synthetase long-chain family member 4 (ACSL4) in the occurrence and development of these diseases. In addition, the article summarizes the application status of ferroptosis models in oral diseases and the current problems faced in the practical application of ferroptosis in dentistry, and proposes strategies to improve therapeutic efficacy, such as developing natural compounds, nano-delivery, and immunocombination therapy. It provides a theoretical basis and practical reference for exploring more targeted therapies through the ferroptosis pathway.
