摘要:铜作为人体必需的微量元素,其稳态失衡引发的铜死亡是近年来新发现的一种细胞死亡方式。铜死亡通过干扰三羧酸循环相关酶系的硫辛酰化结构域,导致毒性蛋白聚集和线粒体应激,与动脉粥样硬化、类风湿关节炎、骨关节炎、炎症性肠病等多种炎症性疾病的发生发展密切相关。随着对铜死亡机制认识的深入,中医药以其多成分、多靶点的调控优势,在干预铜死亡相关炎症反应方面展现出独特潜力。本文系统梳理铜死亡在各类炎症性疾病中的作用机制,并总结中医药通过调节铜稳态、抑制氧化应激及免疫炎症等途径治疗相关疾病的研究进展,旨在为从铜死亡角度阐释中医药作用机制提供理论依据,也为炎症性疾病的临床治疗和药物开发提供新思路。
Abstract: Copper is an essential trace element, and its homeostatic imbalance can induce cuproptosis, a novel form of cell death discovered in recent years. Cuproptosis, primarily by interfering with the lipoylated components of the tricarboxylic acid (TCA) cycle, leads to toxic protein aggregation and mitochondrial stress. It has been closely implicated in the pathogenesis of various inflammatory diseases, including atherosclerosis, rheumatoid arthritis, osteoarthritis, and inflammatory bowel disease. As understanding of the cuproptosis mechanism deepens, Traditional Chinese Medicine (TCM), leveraging its multi-component and multi-target advantages, demonstrates unique potential in mitigating cuproptosis-related inflammatory responses. This article systematically reviews the mechanisms of cuproptosis in various inflammatory diseases and summarizes recent research progress on TCM interventions that modulate copper homeostasis, inhibit oxidative stress, and regulate immune inflammation. The review aims to provide a theoretical basis for elucidating TCMs mechanisms from the perspective of cuproptosis and to offer new insights and potential therapeutic targets for the clinical management of inflammatory diseases.
