    Abstract: Tuberculosis (TB)is a chronic respiratory infectious disease caused by the infection of Mycobacterium tuberculosis (M.tb), which remains one of the deadliest diseases with a significant global threat, severely endangering human life and health. Among the mycobacteria, there exists a special protein family known as the PE/PPE protein family, consisting of two subfamilies, PE and PPE, whose encoding genes occupy 10% of the entire genome. In recent years, many experimental data have fully confirmed that the PE/PPE protein family plays a key role in the pathogenesis of tuberculosis. Therefore, in order to reduce the infection rate and incidence of TB and achieve a zero mortality rate from TB, further research on PE/PPE family proteins is essential. This article will review the structure, antigenicity, physiological functions, and clinical applications of the PE/PPE family genes of Mycobacterium tuberculosis, hoping to provide useful references for the development of immune-enhancing vaccines and serological diagnosis of tuberculosis.
