Causal relationship between 25-hydroxyvitamin D and ankylosing spondylitis: a two-way Mendelian randomization study

    Abstract: To explore whether there is a causal relationship between 25-hydroxyvitamin D (25(OH)D) and AS.Methods:Extract genetic data related to 25(OH)D and AS from genome-wide association studies (GWAS). Employ inverse variance weighting (IVW) for accurate estimation results. Additionally, utilize the MR-Egger regression method, weighted median (WM), and both simple mode and weighted mode for model supplementation. Finally, conduct sensitivity analyses and verification.Results:There is a causal relationship between 25(OH)D levels and the results of the inverse variance weighted (IVW) method in AS, as indicated by an odds ratio (OR) of 0.805 (95% CI: 0.686 – 0.944, P = 0.008). Similarly, the penalised weighted median method yielded consistent results with an OR of 0.799 (95% CI: 0.678 – 0.940, P = 0.007). However, the IVW methods result for AS concerning 25(OH)D indicates no causal relationship, with an OR of 1.019 (95% CI: 0.995 – 1.043, P = 0.110). Furthermore, analyses using the MR-Egger intercept, Cochrans Q test, the Leave-one-out method, and MR-PRESSO revealed no evidence of horizontal pleiotropy or heterogeneity.Conclusion:There may be a genetic causal relationship between 25-hydroxyvitamin D (25(OH)D) concentration and the onset of AS. However, AS does not influence the concentration of 25(OH)D in the body.
